研究概述

  阿尔茨海默病(Alzheimer’s disease,AD)是最常见的神经系统退行性疾病。我国现有AD患者约1000万人,世界第一,且每年仍有180万新增病例,给患者个人、家庭和社会带来严重负担。本课题组长期从AD统计遗传学研究,以AD遗传因素和环境因素为主要研究方向,以“遗传变异—易感基因—生物学通路—调控机制—环境因素”为主线,取得创新性进展(均在国内完成):①在AD遗传变异层面,发现了AD遗传变异,提出并阐明群体遗传异质性和样本量是影响AD遗传变异可重复性的两种重要因素;②在AD易感基因和生物学通路层面,克服传统单点遗传变异分析的局限性,基于遗传变异的相互作用,识别了AD风险基因,采用基因集富集分析,发现了AD相关的神经、免疫、心血管病等生物学通路,建立了AD遗传学研究的“遗传变异—基因—通路”模式;③在“遗传变异—基因—通路”相互作用层面,揭示了AD遗传变异调控基因表达、影响生物学通路,进而引起AD风险的潜在作用机制,且发现这种调控作用具有组织类型和疾病状态的特异性、疾病风险的多效性;④在“遗传—环境”相互作用层面,基于遗传关联研究和孟德尔随机化设计,评估了AD遗传-环境之间的相互作用,阐明了影响AD的社会、病理、生理环境。

Alzheimer's disease (AD) is the most common neurodegenerative disease. The applicant has been engaged in AD statistical genetics research for a long time. First, the applicant first identified novel AD genetic variants and clarified the repeatability mechanisms in different populations at the genome level. Then the applicant investigated the interaction of genetic variants, and identified novel AD susceptibility genes, and neurological, immune, cardiovascular and other biological pathways, which expand the pathogenesis of AD. On this basis, the applicant further integrated the genome and transcriptome data, revealed the underlying mechanism of AD genetic variants regulating gene expression, affecting the biological pathways, and thereby changing the AD risk. Finally, the applicant identified the social, pathological and physiological environments affecting AD using the interaction between genetics and environments.
